CrucialdeterminantsofgeneexpressionpatternsareDNA-bindingtranscriptionfactorsthatchoosegenesfortranscriptional activationorrepressionbyrecognizingthesequenceofDNAbasesintheirpromoterregions.Interactionofthesefactors withtheircognatesequencestriggersachainofevents,ofteninvolvingchangesinthestructureofchromatin,thatleads totheassemblyofanactivetranscriptioncomplex(e.g.,Cosmaetal.1999).Butthetypesoftranscriptionfactorspresentinacellarenotalonesufficienttodefineitsspectrumofgeneactivity, asthetranscriptionalpotentialofagenomecanbecomerestrictedinastablemannerduringdevelopment.Theconstraints imposedbydevelopmentalhistoryprobablyaccountfortheverylowefficiencyofcloninganimalsfromthenucleiofdifferentiated cells(Rideoutetal.2001;WakayamaandYanagimachi2001).A“transcriptionfactorsonly”modelwouldpredictthatthegeneexpressionpatternofadifferentiatednucleuswouldbe completelyreversibleuponexposuretoanewspectrumoffactors.Althoughmanyaspectsofexpressioncanbereprogrammed inthisway(Gurdon1999),somemarksofdifferentiationareevidentlysostablethatimmersioninanaliencytoplasmcannoterasethememory.
